[ Keratan Sulfate specificity: Cross-reactivity with various GAGs ]
We have verified the reactivity of the High Sensitive Keratan Sulfate ELISA Kit with various glycosaminoglycans (GAGs).
Method
Solutions (1,000 ng/mL) of the GAGs listed in the table below were used to prepare a 1/4 dilution series of seven different concentrations (1,000, 250, 62.5, 15.63, 3.91, 0.98, 0.24 ng/mL), and measurements were carried out in accordance with the protocol. The resulting absorbances were shown on a graph.

Results
Conclusion
We have confirmed that the High Sensitive Keratan Sulfate ELISA Kit reacts with Keratan Sulfate derived from bovine cornea (KSBC) and Keratan Polysulfate derived from shark cartilage (KPS), but does not react with Hyaluronic Acid, Chondroitin, Chondroitin Sulfate A, or Heparan Sulfate even at high concentrations of 1,000 ng/mL. Some reaction was observed with Chondroitin Sulfate B, C, D, and E and with Heparin at high concentrations of 1,000 ng/mL. As these reactions were extremely weak at around 1/1,000 of the intensity of the reaction with Keratan Sulfate, however, it has been confirmed that this kit is unaffected by related substances and specifically detects Keratan Sulfate.
